Quality Statement:
Thrombomodulin (TM) is a transmembrane glycoprotein located on theluminal surface of endothelial cells in most normal blood vessels thatregulates coagulation and inflammation. Encoded by an intronless gene on chromosome 20, the mature single-chainTM glycoprotein in the human is 557 amino acids long, structurally organizedinto five distinct regions, including the cytoplasmic and transmembranedomains, a serine/threonine-rich region with an attached chondroitin sulfatemoiety, six consecutive EGF-like repeats, and the N-terminal lectin-likedomain. Thrombomodulin is an endothelial cell thrombin receptor that convertsthrombin into an anticoagulant capable of activating protein C, suggesting aprotective role in thrombosis. TM may also affect astrocyte function andprotect against thrombin-induced neuronal cell death via an APC-dependentmechanism.
